Missions
The PCANS platform (Chromatography and Sugar Analysis Platform) is dedicated to the characterization, structural analysis and purification of glycans. It provides technical and methodological support to the 5 research teams of Cermav. It can also be requested in the context of services (one-off services or service contracts) or research collaboration, for private companies, public research laboratories (CNRS, Universities, INRAE, INSERM, etc.) or other entities requiring its expertise (Hospitals, private laboratories, etc.).
Examples of analysis for services
- Colorimetric determination of total sugar (Dubois method)
- Colorimetric determination of proteins (Bradford method)
- Colorimetric determination of sulfatesdes (Azure A method)
- Qualitative and quantitative analysis of monosaccharides by Ion Chromatography coupled to a Pulsed Amperometric Detector (HPAEC-PAD), or by High Performance Liquid Chromatography coupled to a refractometer (HPLC/RI)
- Qualitative and quantitative analysis of monosaccharides constituting of oligo and polysaccharides, after chemical derivatization, by Gas Chromatography (GC/FID)
- Determination of the osidic bonds of oligo and polysaccharides, after chemical derivation, by Gas Chromatography coupled to Mass Spectrometry (GC/MS)
- Determination of molecular weights and polydispersity index by Steric Exclusion Chromatography coupled with triple detection, UV-Visible, Multi-Angle Light Scattering and Refractometry (HPSEC/UV-MALS-RI)
- Separation of compounds by High Performance Liquid Chromatography coupled with different detectors, Refractometry, UV-Visible or Evaporative Light Scattering Detector (RI-UV or DEDL)
- Fractionation and purification of compounds by Steric Exclusion Chromatography coupled to a Refractometer (SEC/RI)
- Desalting and/or Ultrafiltration of oligo and polysaccharides
Staff
The platform gathers the scientific and technical expertise of the following :
Laurine BUON
Assistant Engineer; specialist in molecular weight analysis by HPSEC/UV-MALS-RI liquid chromatography, and in charge of equipment maintenance;
Eric BAYMA,
Assistant Ingénieur, spécialiste en analyses de poids moléculaires par chromatographie liquide HPSEC/UV-MALS-RI, et responsable de la maintenance des équipements ;
Clara BILLET,
Assistant Engineer, specialized in osidic composition analysis by gas chromatography and ion chromatography.
Equipments
To carry out its missions, PCANS has an unique range of equipment for liquid and gas chromatography.
Liquid Chromatography (10 equipments ):
3 High Pressure Liquid Chromatographs (Ultimate 3000 -ThermoFisher), coupled with different types of detectors (UV/Visible, Refractometer, Evaporative Light Scattering Detector) and equipped with different types of analytical or semi-preparative columns, normal phase, reverse phase, grafted and gel filtration.
Terms of use and access
The platform is open to both the public and private scientific community, within the limits of its expertise, its fields of application and its human and technical resources. The services offered are detailed in the paragraph “Examples of analysis for services”.
The requests or interactions may be carried out in the form of punctual services, service contracts, research contracts or collaborations, depending on the context and the objectives.
How PCANS works
All requests are discussed in advance with the members of the team, either by telephone or by e-mail. Depending on the complexity of the request, a meeting may be considered.
